Experimental study on the effect of Hepatocyte Growth factor for the Liver Regeneration after CCI4 acute intoxication in Rabbits


Abstract
Heptocyte factor(HGF) is the most potent mitogen to mature hepatocytes and stimulates their DNA synthesis in experimental animals as well as in well as in humans. Author tried to assess the effect of HGF on liver regeneration after acute CCI4
intoxication in rabbits.
Sixty-five healthy adult New Zealand White rabbits weighing about 2Kg were divided into three groups: control group (n=5), Acute CCl4 intoxicated group (CCl4 group: n=30), and HGF administered group after CCl4 acute intoxication (HGF group:
n=30).
Five
rabbits were sacrificed each time at 1 day, 3days, 14days, 21days, and 28days after acute CCl4 intoxacation in both CCL4 group and HGF group.
The liver function test, H-E stains, PAS stains, and Ag-NOR stains were carried out in all the groups for evaluation about the effects of HGF after acute CCl4 intoxication.
@ES The results obtained were summerized as follows:
@EN 1. According to histologic findings, swelling of hepatocyte and hydrophic changes in CCl4 group reached peak at 1~3days and returned to control level at 7days. But degree of swollen cell and hydrophic degeneration was lesser in HGF group than
CCl4
group. The mitotic figures, dark cells and double nucleated cells in CCl4 group reached peak at 3 days after CCl4 intoxication and returned to control level at 21~28days after CCl4 intoxication. But degree of mitotic figures, darks cells and
double
nucleated cells was higher in HGF group than in CCl4 group.
2. The degree of positive PAS reation in both CCl4 and HGF grop decreased notably during the period from 1day to 7days and became comparable to control level at 14days after CCl4 intoxication showing no differnces between the two groups.
3. The man value of Ag-NOR index (No. of NOR/100 hepatocytes) of the control group was 165.0¡¾9.5. But in CCl4 group, it was 226.7¡¾18.8 at 1day, 269.5¡¾13.1 at 3days reaching its peak, and returned to control level at 21days after CCl4
intoxication.
In HGF group it reached its peak level of 328.6¡¾18.8, decreased slowly, and reached control level 28days after CCl4 intoxication. The degree of Ag-NRO index was higher and more persistent in HGF than in CCl4 group.
4. In term of AST and ALT changes, their mean values in CCl4 group reached the peak level 1day and returned to control level at 7days after CCl4 intoxication. In HGF group, the mean values of AST and ALT were significantly lower than those in
CCl4
group. But there were no diffences in ALP level between CCl4 and HGF group.
